Specificity
Clone 1M13 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects mitochondrial UMP-CMP kinase 2 (CMPK2). It targets an epitope within 25 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human mitochondrial UMP-CMP kinase 2 (CMPK2).
Application
Quality Control Testing
Evaluated by Western Blotting in THP-1 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CMPK2 in THP-1 cell lysate.
Tested applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected CMPK2 in K562 cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected CMPK2 in human small intestine tissue sections.
Flow Cytometry Analysis: 0.1 μg from a representative lot detected CMPK2 in one million K562 and THP-1 cells.

Target description
UMP-CMP kinase 2, mitochondrial (UniProt: Q5EBM0; also known as EC:2.7.4.14, Cytidine/uridine monophosphate kinase 2, Nucleoside-diphosphate kinase) is encoded by the CMPK2 gene (Gene ID: 129607) in human. UMP-CMP kinase 2 is a mitochondrial enzyme that belongs to thymidine kinase family. It is synthesized with a transit peptide (aa 1-98), which is subsequently cleaved off to produce the mature form (aa 99-449). Its C-terminal domain contains all consensus motifs of a nucleoside monophosphate kinase. It participates in the synthesis of dUTP and dCTP in mitochondria. It phosphorylates dUMP, dCMP, CMP, and UMP with ATP as phosphate donor. However, its kinetic properties differ from cytosolic UMP-CMPK. It displays greatest efficacy to convert dUMP, followed by dCMP. CMP and UMP are relatively poor substrates. Its expression has been detected in chronic myelogenous leukemia K-562 and lymphoblastic leukemia MOLT-4 cells. Its high expression in leukemia cells, dominant expression in bone marrow, and tight correlation with macrophage activation and inflammatory response suggest that it may have other functions in addition to the supply of substrates for mtDNA synthesis. Four isoforms of UMP-CMP kinase 2 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
