General description
Activator Reagent 0.3M 5-benzylmercaptotetrazole (BMT) in acetonitrile (AcN) is also known as benzylthiotetrazole (BTT) or benzyl-1H-tetrazole). It is used as a phosphoramidite activator reagent for oligonucleotide synthesis.
Application
0.3M BMT in AcN can be used as a reagent in solid phase phosphorylation of the oligomers. The activator reacts with the amidite group to form a highly reactive intermediate. The intermediate then forms an internucleotide bond with the 5′hydroxyl group of the growing oligonucleotide chain. The high activation potential of this reagent may be due to its greater acidity (pKa=4.08), which makes it easier to protonate the trivalent phosphorus in the initial stage of activation and increase the speed of the coupling reaction.
