General description
The gene encoding arachidonate 5-lipoxygenase (ALOX5) is localized on human chromosome 10q11.21 and has 13 introns. It spans a region of 71.9 kbp. The ALOX5 protein consists of 673 amino acids and has a molecular weight of 77.9 kDa. It possesses two domains, a regulatory C2-like domain that binds to calcium and a carboxy-terminal catalytic domain. ALOX5 is expressed in B cells, dendritic cells, mast cells, macrophages and polymorphonuclear leukocytes.
This gene encodes a member of the lipoxygenase gene family and plays a dual role in the synthesis of leukotrienes from arachidonic acid. The encoded protein, which is expressed specifically in bone marrow-derived cells, catalyzes the conversion of arachidonic acid to 5(S)-hydroperoxy-6-trans-8,11,14-cis-eicosatetraenoic acid, and further to the allylic epoxide 5(S)-trans-7,9-trans-11,14-cis-eicosatetrenoic acid (leukotriene A4). Leukotrienes are important mediators of a number of inflammatory and allergic conditions. Mutations in the promoter region of this gene lead to a diminished response to antileukotriene drugs used in the treatment of asthma and may also be associated with atherosclerosis and several cancers. The gene ALOX5 (arachidonate 5-lipoxygenase) encodes a lipoxygenase that catalyzes the conversion of arachidonic acid (AA) and other fatty acids to bioactive compounds such as 5(S)-hydroxyeicosatetraenoic acid (5-HETE), leukotriene B4 (LTB4), and other leukotrienes. These leukotrienes and lipoxins function as mediators of inflammatory response. ALOX5 has been implicated in tumorigenesis and may serve as a potential therapeutic target and biomarker.
