General description

Envelope glycoprotein B (UniProt: P13201; also known as gB) is encoded by the gB (also known as UL55) gene (Gene ID: 3077424) in human cytomegalovirus (HHV-5). Human cytomegalovirus (HCMV), a member of the β-Herpesviridae family, is a major cause of morbidity and mortality in organ transplant recipients and patients with AIDS. Its envelope glycoprotein B is reported to be essential for the infectivity and is considered to be one of the most immunogenic virus-encoded proteins. It facilitates the entry of HCMV entry into cells by binding receptors and mediating membrane fusion. Glycoprotein B is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature protein that contains a virion surface domain (aa 23-751), a transmembrane domain (aa 752-772), and an intra-virion domain (aa 773-907). The amino acid sequences 152-158 and 237-244 are critical for fusion and/or binding to host cell membrane. During virion morphogenesis, envelope glycoprotein B probably accumulates in the endosomes and trans-Golgi where secondary envelopment occurs. It is then transported to the cell surface from where it is endocytosed and is directed to the trans-Golgi network. The ectodomain of HCMV glycoprotein B forms rosette-like aggregates that are similar to rosettes formed by the EBV glycoprotein B ectodomain. Four exposed hydrophobic residues in its fusion loop are shown to be essential for rosette formation. A linear domain of glycoprotein B, designated at AD-1, represents a dominant antibody binding site on this protein and is also required for oligomerization of glycoprotein B. Specificity

Clone 27-156 is a mouse monoclonal antibody that detects the envelope glycoprotein B in Human cytomegalovirus (HCMV). It targets an epitope within the antigenic domain 1 (AD1) within the C-terminal half.

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
