Specificity
Clone 2D22 ZooMAb is a rabbit recombinant monoclonal antibody that specifically detects AMPK alpha1. It targets an epitope within 17 amino acids from the C-terminal region.
Immunogen
KLH-conjugated liear peptide corresponding to 17 amino acids from the C-terminal half of human AMPK alpha, isoform 1.
Application
Anti-AMPK alpha, clone 2D22 ZooMAb , Cat. No. ZRB1100, is a recombinant rabbit monoclonal antibody that specifically detects AMPK alpha and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected AMPK alpha in human small intestine and human ovarian cancer tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected AMPK alpha in HeLa cells.

Target description
5′-AMP-activated protein kinase catalytic subunit alpha-1 (UniProt: Q13131; also known as EC:2.7.11.1; AMPK subunit alpha-1, Acetyl-CoA carboxylase kinase, ACACA kinase, Hydroxymethylglutaryl-CoA reductase kinase, HMGCR kinase, Tau-protein kinase PRKAA1) is encoded by the PRKAA1 (also known as AMPK1) gene (Gene ID: 5562) in human. AMPK is a heterotrimer composed of an alpha catalytic subunit (PRKAA1 or PRKAA2), a beta (PRKAB1 or PRKAB2), and a gamma non-catalytic subunit (PRKAG1, PRKAG2 or PRKAG3). Its kinase domain is localized to amino acids 27-279. It also contains an autoinhibitory sequence (aa 302-381) that represses its kinase activity. AMPK is an energy sensor protein kinase that plays a key role in regulating cellular energy metabolism. In response to reduction of intracellular ATP levels, AMPK activates energy-producing pathways and inhibits energy-consuming processes, including inhibition of protein, carbohydrate and lipid biosynthesis, and reduces cell growth and proliferation. AMPK acts via direct phosphorylation of metabolic enzymes, and by longer-term effects via phosphorylation of transcription regulators. AMPK is shown to regulate insulin-signaling and glycolysis by phosphorylating IRS1, PFKFB2 and PFKFB3 and it stimulates glucose uptake in muscle by increasing the translocation of the glucose transporter GLUT4 to the plasma membrane. In the liver it acts as a key regulator of glucose homeostasis by phosphorylating CRTC2/TORC2, leading to CRTC2/TORC2 sequestration in the cytoplasm. AMPK is activated by phosphorylation on threonine 183 by STK11/LKB1. Binding of AMP to non-catalytic gamma subunit can result in allosteric activation, inducing phosphorylation on threonine 183. AMP-binding to gamma subunit also sustains activity by preventing dephosphorylation of threonine 183. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose. Normal appearance is a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
