General description
Adenomatous polyposis coli protein (UniProt P25054; also known as Deleted in polyposis 2.5, Protein APC) is encoded by the APC (also known as DP2.5) gene (Gene ID 324) in human. The multifunctional tumor suppressor APC is a large scaffold protein that contains multiple armadillo repeat (ARM) domains (a.a. 453-767 of human APC spliced isoform 1) for mediating various cellular functions, including Wnt signal regulation, cell adhesion, and chromosomal segregation during mitosis. APC, AXIN, casein kinase 1 (CK1) and glycogen synthase kinase 3 (GSK3) constitute the core proteins of the β-catenin destruction complex that functions as the negative regulator against Wnt signaling transduction. Mice with Apc gene mutations develop adenomas in the intestine due to Wnt hyperactivation, and functional loss of APC due to gene mutations is the hallmark of human colorectal cancers (CRC). APC also directs cholinergic synapse assembly between neurons and is commonly employed as a marker for monitoring stem cells neuronal differentiation.

Immunofluorescence Analysis: A representative lot detected a granular APC staining pattern at a subapical region of epithelial cells in fallopian mucosa by fluorescent immunohistochemistry staining of paraffin-embedded, paraformaldehyd-fixed fallopian tube tissue sections (Kessler, M., et al. (2012). Am. J. Pathol. 180(1):186-198).
