Specificity

Clone 1H2 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Aryl hydrocarbon receptor nuclear translocator (ARNT). It targets an epitope within 17 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half of human Aryl hydrocarbon receptor nuclear translocator (ARNT).

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ARNT in NIH3T3 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ARNT in L6 cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ARNT in MCF-7 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ARNT in human uterus tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound ARNT with a KD of 1.0 x 10-12 in an affinity binding assay.

Target description

Aryl hydrocarbon receptor nuclear translocator (UniProt: P27540; also known as ARNT protein, Class E basic helix-loop-helix protein 2, bHLHe2, Dioxin receptor, nuclear translocator, Hypoxia-inducible factor 1-beta, HIF-1-beta, HIF1-beta) is encoded by the ARNT (also known as BHLHE2) gene (Gene ID: 405) in human. ARNT is a member of the basic Helix-Loop-Helix PER/ARNT/SIM (bHLH/PAS) protein family and is conserved as an important transcriptional regulator in organogenesis, neural development, and physiological adaptation to hypoxia. It exhibits about 90% amino acid identity with ARNT2, but is more ubiquitous in its distribution in adult tissues. It exists as a monomer, but dimerizes with Aryl Hydrocarbon Receptor (AhR) to form a heterodimer that binds to core DNA sequence 5′-TGCGTG-3′ within the dioxin response element (DRE) of target gene promoters and activates their transcription. ARNT also forms a heterodimer with HIF-1a and that dimer promotes cell survival and angiogenesis at varying oxygen levels. It facilitates normoxia under adverse conditions. ARNT contains one bHLH domain (aa 89-142), two PAS (Per-Arnt-Sim) domains (aa 161-235 and 349-419) and one PAC domain (aa 424-467). Despite the presence of a nuclear export sequence, it mainly accumulates in the nucleus. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
