Specificity
Clone 1B15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects β-Arrestin 1. It targets an epitope within 22 amino acids from the C-terminal region.

Application
Quality Control Testing
Evaluated by Western Blotting in MCF-7 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected β-Arrestin 1 in MCF-7 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected β Arrestin 1 in Human Lung, T47D, Raw264.7, and Rat Lung lysates.

Target description
Beta-Arrestin-1 (UniProt:P49407; also known as Arrestin beta-1, Non-visual arresrtin-2) is encoded by the ARRB1 (also known as ARR1) gene (Gene ID: 408) in human. Arrestins are a superfamily of multi-functional proteins that regulate signaling and trafficking of the majority of G-protein-coupled receptors (GPCRs), as well as sub-cellular localization and activity of many other signaling proteins. β-Arrestin can exist as a monomer or as a homodimer. Its self-association is mediated by InsP6-binding. It can also form homooligomer with β-arrestin 2 (ARRB2) that is mediated by InsP6-binding. It is detected on cytoplasm, nucleus, and membrane. The monomeric form is predominantly located in the nucleus and the oligomeric form is found in the cytoplasm. It translocates to the nucleus upon stimulation of opioid receptor 1 (OPRD1). β-arrestin 1 is reported to play a role in regulating agonist-mediated GPCR signaling by mediating both receptor desensitization and re-sensitization processes. It also targets many receptors for internalization by acting as endocytic adapters and recruiting the GPRCs to the adapter protein 2 complex 2 (AP-2) in clathrin-coated pits. However, its involvement appears to vary significantly depending on the receptor, agonist, and cell type. Internalized arrestin-receptor complexes are shown to traffic to intracellular endosomes, where they remain uncoupled from G-proteins. Two different modes of arrestin-mediated internalization have been reported. Class A receptors, such as adrenoceptor β2 and opioid receptor 1, dissociate from β-arrestin at or near the plasma membrane and undergo rapid recycling. However, class B receptors, such as arginine-vasopr+D5essin receptor 2 and neurotensin receptor 1, internalize as a complex with arrestin and traffic with it to endosomal vesicles, presumably as desensitized receptors, for extended periods of time. β-arrestin 1 is constitutively phosphorylated at serine 412 in the cytoplasm. At the plasma membrane, is rapidly dephosphorylated, a process that is required for clathrin binding and adrenoceptor β2 endocytosis but not for adrenoceptor β2 binding and desensitization. Once internalized, it is re-phosphorylated. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
