Specificity
Clone 1G13 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Sarcoplasmic/endoplasmic reticulum calcium ATPase 2 (SERCA2). It targets an epitope within 17 amino acids from the N-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal region of human Sarcoplasmic/endoplasmic reticulum calcium ATPase 2.
Application
Quality Control Testing
Evaluated by Western Blotting in A549 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ATP2A2/SERCA2 in A549 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ATP2A2/SERCA2 in C2C12 cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ATP2A2/SERCA2 in Human skeletal muscle tissue sections.
Affinity Binding Assay:: A representative lot of this antibody bound ATP2A2/SERCA2 peptide with a KD of 3.6 x 10-7 in an affinity binding assay.

Target description
Sarcoplasmic/endoplasmic reticulum calcium ATPase 2 (UniProt: P16615; also known as EC:7.2.2.10, SERCA2, SR Ca(2+)-ATPase 2, Calcium pump 2, Calcium-transporting ATPase sarcoplasmic reticulum type slow twitch skeletal muscle isoform, Endoplasmic reticulum class 1/2 Ca(2+) ATPase) is encoded by the ATP2A2 (also known as ATP2B) gene (Gene ID: 488) in human. ATP2A2/SERCA2 is a muti-pass membrane protein with six cytoplasmic domains, ten transmembrane domains, and five lumenal domains. It serves as a magnesium-dependent enzyme that catalyzes the hydrolysis of ATP coupled with the translocation of calcium from the cytosol to the sarcoplasmic reticulum lumen. Five isoforms of ATP2A2/SERCA2 have been described that are produced by alternative splicing. Isoform 1 is widely expressed in smooth muscle and non-muscle tissues with highest expression in liver, pancreas, and lung. Isoforms 2 and 3 are highly expressed in the heart and slow twitch skeletal muscle. ATP2A2/SERCA2 exhibits different conformational states with differential Ca2+ affinities. The E1 conformational state (active form) shows high Ca2+ Affinity™, while the E2 state exhibits low Ca2+ Affinity™. Ca2+ binding to the cytosolic domain of the pump in the high-Affinity™ E1 conformation is followed by the ATP-dependent phosphorylation of the active site Asp, giving rise to E1P. A conformational change of the phosphoenzyme gives rise to the low-Affinity™ E2P state that exposes the Ca2+ ions to the lumenal side and promotes Ca2+ release. Dephosphorylation of the active site Asp mediates the subsequent return to the E1 conformation. Under conditions of oxidative stress, it undergoes nitration on two tyrosine residues (aa 294, 295) that reduces its catalytic activity. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
