Specificity

Clone 1C6 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects BRG1-associated factor 57 (BAF57/SMARC1). It targets an epitope within 23 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the C-terminal region of human BRG1-associated factor 57 (BAF57/SMARC1).

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected BAF57/SMARCE1 in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected BAF57/SMARCE1 in NIH3T3 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected BAF57/SMARCE1 in human cerebral cortex tissue sections.

Affinity Binding Assay Analysis: A representative lot of this antibody bound BAF57-SMARCE1 with a KD of 9.4 x 10^-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected BAF57/SMARCE1 in MCF-7 cells.

Target description

SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily E member 1 (UniProt: Q969G3;  also known as BRG1-associated factor 57, BAF57) is encoded by the BAF57 (also known as SMARCE1) gene (Gene ID: 6605) in human. BAF57 is a nuclear protein that is a component of the multiprotein chromatin-remodeling complexes SWI/SNF: SWI/SNF-A (BAF), SWI/SNF-B (PBAF) and related complexes. It is involved in transcriptional activation and repression of select genes by chromatin remodeling. The main structural feature of BAF57 is a high-mobility group domain (aa 66-134), the DNA-binding properties of which suggest that BAF57 has topological roles as the BAF complex enters or exits the nucleosome. The HMG domain is essential for CD4 silencing and CD8 activation. BAF57 displays specific interactions with a number of proteins outside the BAF complex through which  it can accomplish specific functions. During embryonic development, BAF57 is responsible for the silencing of the CD4 gene during T-cell differentiation, and during the repression of neuronal genes in non-neuronal cells, BAF57 interacts with the transcriptional corepressor, Co-REST, and facilitates repression. BAF57 belongs to the neural progenitors-specific chromatin remodeling complex (npBAF complex) and the neuron-specific chromatin remodeling complex (nBAF complex). During neural development a switch from a stem/progenitor to a post-mitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. Mutations in BAF57 gene are known to cause Meningioma, a common neoplasm of the central nervous system derived from arachnoidal cells. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
