Specificity
Clone 1E9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects B-cell lymphoma 3 protein (Bcl3). It targets an epitope within 19 amino acids from the N-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal region of human B-cell lymphoma 3 protein (Bcl3).
Application
Quality Control Testing
Evaluated by Western Blotting with GST-tagged, recombinant Human Bcl3.
Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected GST-tagged, recombinant Human Bcl3.
Tested Applications
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Bcl3 in HeLa, A431, and HepG2 cells.
Affinity Binding Assay: A representative lot of this antibody bound Bcl3 peptide with a KD of 1.4 x 10-7 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Bcl3 in human placenta tissue sections.

Target description
B-cell lymphoma 3 protein (UniProt: P20749; also known as Bcl3, Proto-oncogene BCL3) is encoded by the BCL3 (also known as D19S37) gene (Gene ID: 602) in human. Bcl3, a nuclear transcription factor, is a member of the I B family of NF- B inhibitors. It is localized in the nucleus as well as in cytoplasm. In the cytoplasm, it is shown to inhibit the nuclear translocation of the p50 subunit of NF- B and in the nucleus, it acts as transcriptional activator that promotes transcription of NF- B target genes. Ubiquitination via lysine 63-linked ubiquitin chains is required for its nuclear accumulation. Bcl3 can be phosphorylated by Akt, Erk2, and IKK1/2. Phosphorylation of serine 33 by Akt is reported to induce switching of K48 ubiquitination to K63 ubiquitination that promotes its nuclear localization and stabilization. Phosphorylation of seine 114 and 446 by Erk2 and IKK1/2 converts Bcl3 into a transcriptional coregulator by facilitating its recruitment to DNA. Cells with S114A/S446A mutations exhibit cellular proliferation and migration defects. Bcl3 can also be phosphorylated by GSK3 on serine 394 and serine 398, which leads to its degradation through the proteasome pathway. A chromosomal aberration involving Bcl3 is reported to cause B-cell chronic lymphocytic leukemia (B-CLL). Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
