Specificity
Clone 1G3 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Bid.

Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Bid in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:19,000 dilution from a representative lot detected Bid in lysate from MCF-7 cells and 1:1,000 dilution in lysates from C2C12, and PC12 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Bid in Human tonsil tissue sections.
Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected Bid in Human tonsil tissue sections.

Target description
BH3-interacting domain death agonist (UniProt: P55957; also known as p22 BID, BID) is encoded by the BID gene (Gene ID: 637) in human. Bid is a BH3 domain containing member of the Bcl-2 family with proapoptotic function. It is shown to be crucial for death receptor-mediated apoptosis in many cell systems where it counters the protective effects of Bcl-2. It links the death receptor signaling pathway to the mitochondrial signaling pathway mediated by Bcl-2 proteins. Full length Bid that is localized in the cytoplasm is cleaved by caspase-8 to generate a truncated form (tBid) that translocates to mitochondria. tBid then binds to mitochondrial Bak and induces the release of cytochrome c. TNFα-induced caspase-mediated cleavage of Bid produced p15, 13, and p11 fragments. Caspase-8-mediated Bid processing of Bid bridges the extrinsic and intrinsic apoptosis pathways that provide a mechanism to amplify death signals. The BH3 motif of Bid is localized in amino acids 86-100. The hydrophobic FACE® of the BH3 domain, which is exposed in other family members, is enclosed by the N-terminal of Bid and only exposed following proteolytic cleavage of Bid. A second functional domain in Bid known as the BH3B domain, which is localized in the N-terminus region, is reported to suppress the apoptogenic activity of the BH3 domain. The region between these two opposing domains is sensitive to proteolytic cleavage and contains cleavage sites for caspases and granzyme B. Four isoforms produced by Alternative splicing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
