Specificity

Clone 1K4 is a Rabbit recombinant monoclonal antibody that specifically detects Voltage-dependent calcium channel subunit alpha-2/delta-1 (CACNA2D1). It targets an epitope within 22 amino acids from the N-terminal, extracellular domain.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal, extracellular domain of human Voltage-dependent calcium channel subunit alpha-2/delta-1 (CCNA2D1).

Application

Quality Control Testing

Evaluated by Immunocytochemistry in U-251 MG cells.

Immunocytochemistry Analysis: A 1:100 dilution of this antibody detected CACNA2D1 in U-251 MG cells.

Tested applications

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CACNA2D1 in human skeletal muscle.

Affinity Binding Assay: A representative lot of this antibody bound CACNA2D1 with a KD of 7.9 x 10-7 in an affinity binding assay.

Target description

Voltage-dependent calcium channel subunit alpha-2/delta-1 (UniProt: P54289; also known as Voltage-gated calcium channel subunit alpha-2/delta-1, CACNA2D1) is encoded by the CACNA2D1 (also known as CACNL2A, CCHL2A, MHS3) gene (Gene ID: 781) in human. CACNA2D1 is a dimeric protein formed by disulfide-linked 2 and chains. It regulates calcium current density and activation/inactivation kinetics of the calcium channel and is shown to play an important role in excitation-contraction coupling. It is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 25-1073), a transmembrane domain (aa 1074-1094), and a short cytoplasmic tail (aa 1095-1103). It also has a MIDAS-like motif (aa 259-263), which is present in its VWFA domain (aa 253-430). The MIDAS-like motif is shown to bind divalent metal cations and is required to promote trafficking of the a-1 (CACNA1) subunit to the plasma membrane by an integrin-like switch. Five different isoforms of CACNA2D1 have been described that are produced by alternative splicing. Isoform 1 is expressed in skeletal muscle, isoform 2 is expressed in the central nervous system, and isoforms 3, 4, and 5 are expressed in the aorta. Isoforms 2, 4, and 5 are also observed in neuroblastoma cells. Mice bearing point mutations in the CACNA2D1 gene display reduced response to pregabalin and an abnormal central nervous system synaptic transmission. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
