General description
Voltage-sensitive calcium channels (vscc) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1c gives rise to l-type calcium currents. Long-lasting (l-type) calcium channels belong to the "high-voltage activated" (hva) group. They are blocked by dihydropyridines (dhp), phenylalkylamines, benzothiazepines, and by omega-agatoxin-IIIA (omega-aga-IIIA). They are however insensitive to omega-conotoxin- gvia (omega-ctx-gvia) and omega-agatoxin-IVA (omega-aga-IVA) (Swiss Prot).
Specificity
Recognizes all forms of alpha 1D subunit from the voltage-gated calcium channel. Does not cross react with any other calcium channel antigens tested so far. Sequence conservation: hamster and mouse (16/17); human (15/17); chicken (12/17) residues identical.
Immunogen
Purified peptide CND1 from 859-875 of alpha 1D subunit of rat brain voltage-gated calcium channel (Accession P27732).
Application
Anti-Calcium Channel Antibody, Voltage Gated α 1D is an antibody against Calcium Channel for use in IH, IP & WB.
In preparing full-length protein samples for Western blot or immunoprecipitation, the procedures should be performed at 4°C, with the following protease inhibitor mixture: pepstatin A (1 μg/mL), leupeptin (1 μg/mL), aprotinin (1 μg/mL), Pefabloc SC (0.2 mM), benzamidine (0.1 mg/mL), and calpain inhibitors I and II (8 μg/mL each).
Western blot: 1:200 using ECL on rat membrane preperations. LMW form shows a 190kDa band while the HMW form shows a 210kDa band.
Immunohistochemistry on rat brain sections.
Dilutions should be made using a carrier protein such as BSA (1-3%).
Optimal working dilutions and protocols must be determined by the end user.

Physical form
Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, 5% sucrose as a stabilizer and 0.05% sodium azide as a preservative. Reconstitute with 200 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).
Storage and Stability
Maintain lyophilized material at -20°C for up to 12 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Control
CONTROL ANTIGEN: Included free of charge with the antibody is 20 μg of control antigen (lyophilized powder in phosphate buffered saline, pH 7.4, containing 5% sucrose and 0.025% sodium azide). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
