Specificity

Clone 1E21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Caspase-7. It targets an epitope within 20 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal region of human Caspase-7.

Application

Quality Control Testing

Evaluated by Western blotting in lysate from Jurkat cells treated with etoposide.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Caspase-7 in lysates from Jurkat cells overnight serum starved and treated with etoposide (100 μM; 2 h), but not in untreated cells.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Caspase-7 in A549 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Caspase-7 in human small intestine tissue sections.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Caspase-7 in one million Jurkat cells treated with etoposide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Target description

Caspase-7 (UniProt: P55210; also known as EC:3.4.22.60, CASP-7, Apoptotic protease Mch-3, CMH-1, ICE-like apoptotic protease 3, ICE-LAP3) is encoded by the CASP7 (also known as MCH3) gene (Gene ID: 840) in human. Activation of caspases is one of the most widely recognized features of apoptosis. Caspases are secreted as latent single chain zymogens, which are further processed to create active caspases that initiate the death program by destroying key components of the cellular infrastructure and activating factors that mediate damage to the cells. An important feature of caspases is that they are not associated with any nonspecific degradative process but are activated only in response to specific signaling event. Caspase-7, a member of the executioner group of caspases, is a cytosolic enzyme that is synthesized as a pro-caspase consisting of two propeptide (aa 2-23 and 199-206) that are cleaved off by granzyme B or caspase-3 to produce the active caspase-7 that consists of a p20 (aa 24-198) and a p11 (aa 207-303) subunits. In the inactive form these two subunits are connected by a linker sequence that hinders ordering of the active site. The removal of this linker region by cleavage of the peptide bond between Asp198 and Ser199 (in p20) and Asp206 and Ala207 (in p11) activates this enzyme. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
