Specificity
Clone 1B21 is a Rabbit recombinant monoclonal antibody that specifically detects Cyclin-dependent kinase 2 (Cdk2). It targets an epitope within 25 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal half of human Cyclin-dependent kinase 2 (Cdk2).
Application
Quality Control Testing
Evaluated by Western Blotting in HEK293 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Cdk2 in HEK293 cell lysate.
Tested applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Cdk2 in lysates from RAW264.7 cells, Rat kidney tissue, and MCF-7 cells.
Affinity Binding Assay: A representative lot of this antibody bound Cdk2 with a KD of 3.7 x 10-8 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Cdk2 in HeLa and NIH 3T3 cells..
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Cdk2 in human tonsil tissue sections.

Target description
Cyclin-dependent kinase 2 (UniProt: P24941;also known as EC:2.7.11.22, Cell division protein kinase 2, p33 protein kinase) is encoded by the CDK2 (also known as CDKN2) gene (Gene ID: 1017) in human. Cdk2 is a multifunctional serine/threonine-protein kinase that is involved in the control of the cell cycle and plays a role in germ cell development. It is localized in the nucleus, cytoplasm, and endosomes. It is considered to be important in orchestrating a fine balance between cellular proliferation, cell death, and DNA repair in human embryonic stem cells. Its activity is reported to be maximum during S phase and G2 phase and it is activated by interaction with cyclin E during the early stages of DNA synthesis to permit G1-S transition. It is subsequently activated by cyclin A2 during the late stages of DNA replication to drive the transition from S phase to G2. Cdk2 phosphorylation at threonine 160 by Cdk7 in a Cdk activating kinase (CAK) complex promotes its kinase activity. However, phosphorylation at threonine 14 and tyrosine 15 during S and G2 phases inactivates the enzyme and dephosphorylation by CDC25A is required for its activation. Active Cdk2 is reported to phosphorylate numerous substrates, including catenin b1, Ubiquitin Specific Peptidase 37 (USP37), p53/TP53, nucleophosmin 1, CDK7, RB1, MYC, and BRCA2. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
