General description
Interleukin-17F (UniProt: Q7TNI7; also known as IL-17F) is encoded by the Il17f gene (Gene ID: 257630) in murine species. The IL-17 family of cytokines contains six members that share four conserved cysteine residues at their C-terminal region that participate in the formation of intermolecular disulfide linkages. Members of this family are associated with the pathogenesis of multiple autoimmune diseases including rheumatoid arthritis, multiple sclerosis, and inflammatory bowel diseases. IL-17F is an effector cytokine of innate and adaptive immune system involved in antimicrobial host defense and maintenance of tissue integrity. It is a disulfide-linked homodimeric protein that is synthesized with a signal peptide (aa 1-28), which is subsequently cleaved off to produce mature secreted protein (aa 29-161). It can also form a heterodimer with IL-17A. Its actions are mediated via interaction with IL-17RA and IL-17RC receptors with 2:1 binding stoichiometry. It is primarily involved in host defense against extracellular bacteria and fungi by inducing neutrophilic inflammation. As signature effector cytokine of T-helper 17 cells (Th17), it induces neutrophil activation and recruitment at infection and inflammatory sites. It is also known to stimulate the production of antimicrobial β-defensins by mucosal epithelial cells thereby limiting the entry of microbes through the epithelial barriers. Specificity
Clone MM17F-8F5 is a mouse monoclonal antibody that detects Interleukin-17F.
Immunogen
Ovalbumin-conjugated, full-length recombinant murine IL-17F.
Application
Quality Control Testing
Evaluated by Western Blotting with recombinant mouse IL-17F.
Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected recombinant mouse IL-17F.
Tested Applications
Flow Cytometry Analysis: A representative lot detected Interleukin-17F in Flow Cytometry application (St. Leger, A.J., et al. (2017). Immunity. 47(1):148-158.e5).
ELISA Analysis: Various dilutions of this antibody detected recombinant mouse IL-17F.
Neutralizing: A representative lot neutralized IL-17F activity and reduced neutrophil chemotactic activity produced by IL-17A / splenocytes activated under Th17 conditions.(Uyttenhove, C., et al. (2011). J Leukoc Biol. 89(6):1001-7).
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
