Immunogen
synthetic peptide corresponding to amino acids 1321-1339 of human claspin, conjugated to KLH via an N-terminal added cysteine residue.
Application
Anti-Claspin (C-terminal) antibody is suitable for microarray and western blot analysis at a dilution of 1:250-1:500.
Biochem/physiol Actions
Claspin is a protein which plays a major role in DNA replication. It involves in protecting DNA replication forks against DNA damage. In DNA damage response, claspin directly interacts with Rad17, which is required for ATR activation. ATR is activated by various DNA damage and DNA replication stress. Once activated, ATR (ataxia telangiectasia mutated-Rad3 related protein) phosphorylates effector kinase Chk1 which further depends on mediator protein claspin. Claspin helps to stimulate phosphorylation reaction of Chk1.
Physical form
Solution in 0.01 M phosÂÂphate buffered saline, pH 7.4, containing 15 mM sodium azide.
