General description
DTX3L (deltex 3 like), also known as BBAP (B-aggressive lymphoma and BAL1 binding partner), is an E3 ubiquitin ligase. This gene localizes to human chromosome 3q21. Its C-terminal shares high identity with deltex (DTX) family members and contains a RING domain.

Application

Biochem/physiol Actions
DTX3L (deltex 3 like) is responsible for controlling the subcellular localization of BAL1 (B-aggressive lymphoma 1) protein in a dynamic shuttling manner. The expressions of both these protein are high in host response diffuse large B-cell lymphoma (HR-DLBCL). This protein regulates DNA damage response by monoubiquitylating histone H4 at lysine 91 position. It functions as a binding partner of BAL and they are both up-regulated in chemoresistant lymphomas. This protein plays a role in the endosomal sorting of CXCR4, which is a GPCR (G-protein couple receptor). Crosstalk amongst proteins STAT1 (signal transducer and activator of transcription 1), DTX3L and ARTD-like mono-ADP-ribosyltransferases is involved in the proliferation and survival of prostate cancer cells. Thus, in prostate cancer of high risk tumors with elevated STAT1 signaling, targeting these proteins can improve the outcome of chemotherapy or radiation therapy.
