Specificity
Clone 1K8 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects excitatory amino acid transporter 2 (EAAT2). It targets an epitope within 20 amino acids from the C-terminal region.

Target description
Excitatory amino acid transporter 2 (UniProt: P43004; also known as Glutamate/aspartate transporter II, Sodium-dependent glutamate/aspartate transporter 2, Solute carrier family 1 member 2, EAAT-2) is encoded by the SLC1A2 (also known as EAAT2, GLT1) gene (Gene ID: 6506) in human. EAAT-2 is a multi-pass homotrimeric membrane protein serves as a major sodium-dependent, high-affinity transporter that moves glutamate back into the cell following neurotransmission. It is expressed predominantly in astroglial cells where it is responsible for about 90% of total glutamate uptake and maintains extracellular glutamate below excitotoxic levels. EAAT-2 functions as a symporter that transports one amino acid molecule together with two or three Na+ ions and one proton, in parallel with the counter-transport of one K+ ion. EAAT-2 contains 6 cytoplasmic domains, 5 extracellular domains, and 8 transmembrane regions plus two helical hairpins that dip into the membrane. These helical hairpin structures play an important role in the transport process. The first enters the membrane from the cytoplasmic side, the second one from the extracellular side. During the transport cycle, the regions involved in amino acid transport, and especially the helical hairpins, move vertically by about 1.5 to 1.8 nm, alternating between exposure to the aqueous phase and reinsertion in the lipid bilayer. In contrast, the regions involved in trimerization do not move. Mutation in SLC1A2 gene can cause early infantile epileptic encephalopathy, a heterogeneous group of severe childhood onset epilepsies characterized by refractory seizures, neurodevelopmental impairment, and poor prognosis. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
