Specificity
Clone 1I18 is a Rabbit recombinant monoclonal antibody that specifically detects F-Box only protein 38 (FBXO38). It targets an epitope within the internal region.
Immunogen
His-tagged recombinant fragment corresponding to 300 amino acids from the internal region of human F-Box only protein 38 (FBXO38).
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected FBXO38 in HeLa cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected FBXO38 in NIH3T3 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound FBXO38 with a KD of 5.0 x 10-9 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected FBXO38 in human colon tissue sections.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected FBXO38 in HeLa and HepG2 cells.

Target description
F-box only protein 38 (UniProt: Q6PIJ6; also known as FBXO38) is encoded by the FBXO38 (also known as SP329) gene (Gene ID: 81545) in human. FBXO38 is a component of the SKP1-CUL1-F-Box (SKF) E3 ubiquitin ligase complex that is reported to mediate lysine 48-linked polyubiquitination and subsequent degradation of PD-1 and thereby regulates T-cell-mediated immunity. Ectopic expression of FBXO38 leads to lower levels of endogenous PD-1 on the cell surface of activated T-cells, and FBXO38 knockdown causes higher levels of PD-1 on cell surface. Its levels are shown to be upregulated by IL-2 and are down-regulate in tumor infiltrating T-cells. It may also indirectly stimulate the activity of transcription factor KLF7, a regulator of neuronal differentiation, without promoting KLF7 ubiquitination. FBXO38 predominantly resides in the cytosol and is exported by the nucleus in a CRM1-dependent manner. It has three nuclear export signals (aa 194-201; 307-316; and 451-460) and one nuclear localization signal (aa 896-899). Its F-Box domain resides in amino acids 30-75. Mutations in FBXO38 gene are known to cause distal hereditary motor neuronopathy that is characterized by onset of slowly progressive distal lower limb weakness and atrophy between the second and fourth decades of life. Three isoforms of FBXO38 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
