Specificity
Clone 1H12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects FOXA2. It targets an epitope within 17 amino acids from the N-terminal half.

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human FOXA2.
Application
Quality Control Testing
Evaluated by Western Blotting in A549 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected FOXA2 in A549 cell lysate.
Tested applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FOXA2 in A549 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected FOXA2 in human colon tissue sections.

Target description
Hepatocyte nuclear factor 3-beta (UniProt: Q9Y261; also known as HNF-3-beta, HNF-3B, Fork head box protein A2, Transcription factor 3B, TCF-3B) is encoded by the FOXA2 (also known as HNF3B, TCF3B) gene (Gene ID: 3170) in human. The FOX family of transcription factors contains more than 50 members, all of which share a winged-helix DNA binding domain. FOXA2 is a fork-head box transcription factor that is involved in embryonic development, establishment of tissue-specific gene expression, and regulation of gene expression in differentiated tissues. It is required for notochord formation and is involved in the development of multiple endoderm-derived organ systems such as the liver, pancreas, and lungs. It contains two transactivation domains (aa 14-93 and 361-457) and its fork-head domain is localized in amino acids 159-252. It shuttles between the nucleus and cytoplasm in a CRM1-dependent manner and in response to insulin signaling via AKT1 it is exported from the nucleus. Phosphorylation of FOXA2 at threonine 156 by AKT inactivates its transcription function and induces its nuclear exclusion. Deletion of FOXA2 in the epithelial cells of the developing mouse lung is shown to inhibit the differentiation of respiratory epithelial cells and causes neonatal respiratory failure, with the characteristics of respiratory distress syndrome in preterm infants. FOXA2 knockdown is reported to induce EMT and increase invasion of A549 lung cancer cells and overexpression of FOXA2 inhibits human lung cancer cell invasion and migration. Two isoforms of FOXA2 have been described that are produced by alternative splicing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
