Specificity
Clone 4J2 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Forkhead box protein P3 (FoxP3). It targets an epitope within 14 amino acids from the C-terminal, the DNA binding domain.
Immunogen
KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal, DNA-binding region of human Forkhead box protein P3.
Application
Anti-FoxP3, clone 4J2 ZooMAb, Cat. No. ZRB1565, is a recombinant Rabbit monoclonal antibody that specifically targets FoxP3 and is tested for use in Affinity Binding Assay, Immunocytochemistry, and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FoxP3 in HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound FoxP3 peptide with a KD of 1.7 x 10-7 in an affinity binding assay.

Target description
Forkhead box protein P3 (UniProt: Q9BZS1; also known as Scurfin, FoxP3) I encoded by the FOXP3 (also known as IPEX, JM2) gene (Gene ID: 50943) in human. FoxP3 is homodimeric protein that Is predominantly expressed in the cytoplasm in activated conventional T-cells whereas it is mainly expressed in the nucleus in regulatory T-cells (Treg). It serves as a transcriptional regulator and is crucial for the development and inhibitory function of regulatory T-cells. The 41 kDa form derived by proteolytic processing is found exclusively in the chromatin fraction of activated Treg cells. Its levels are reported to be down-regulated in regulatory T-cells during inflammation. FoxP3 can undergo proteolytic cleavage in activated regulatory T-cells and can be cleaved at either the N- (aa 51-52) or C-terminal (aa 417-418) site, or at both sites. FoxP3 plays an essential role in maintaining homeostasis of the immune system by allowing the acquisition of full suppressive function and stability of the Treg lineage, and by directly modulating the expansion and function of conventional T-cells. Its dimerization is reported to be essential for its transcriptional regulator activity. Is phosphorylation at serine 418 is shown to regulate its transcriptional repressor activity and consequently, regulatory T-cells suppressive function. FoxP3 is synthesized with a propeptide (aa 418-431), which is subsequently cleaved off in the mature form. Its nuclear localization signal is in the amino acids 414-417 and 68-76 and its nuclear export signal is localized in amino acids 239-249. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
