Specificity

Clone 1L9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects RNA-binding protein FXR1. It targets an epitope within 25 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human RNA-binding protein FXR1.

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected FXR1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected FXR1 in HepG2 cell lysate and 1:10,000 dilution in Human brain tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound FXR1 peptide with a KD of 1.1 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected FXR1 in HepG2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected FXR1 in human cerebral cortex tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

RNA-binding protein FXR1 (UniProt: P51114; also known as FMR1 autosomal homolog 1, hFXR1p) is encoded by the FXR1 gene (Gene ID: 8087) in human. FXR1 is a cytoplasmic protein that is expressed in all tissues examined. It contains RNA-binding domains that are localized in amino acids 222-251 (KH1); 285-314 (KH2) and 442-457 (RNA-binding RGG-box ) and differentially regulate RNA translation and stability. It is required for embryonic and postnatal development of muscle tissue. Three isoforms of FXR1 have been described that are produced by alternative splicing. The isoform balance is reported to be altered in myotonic dystrophy, facioscapulohumeral muscular dystrophy, and in diabetes. It is a muscle-enhanced and IL-19- inducible RNA-binding protein that acts as a negative regulator of inflammatory gene expression by competing with HuR on the 30 UTR of inflammatory mRNA transcripts. It is required for IL-19-dependent reduction of HuR mRNA stability and abundance. FXR1 are also shown to form amyloid oligomers and insoluble aggregates in rat cortical neurons and these amyloid conformers are resistant to RNase treatment.  In its monomeric form,  FXR1 are almost undetectable. FXR1 depletion is shown to be perinatally lethal in mice, Xenopus, and zebrafish.  This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
