General description
Potassium channel subfamily T member 1 (UniProt: Q9Z258; also known as Sequence like a calcium-activated potassium channel subunit, Slack, KCNT1) is encoded by the Kcnt1 (also known as Slack) gene (Gene ID: 60444) in rat. KCNT1 is a multi-pass membrane protein that serves as an outwardly rectifying potassium channel subunit that may co-assemble with other Slo-type channel subunits. It represents the largest known potassium channel subunit. It is activated by high intracellular sodium or chloride levels and is activated upon stimulation of G-protein coupled receptors, such as CHRM1 and GRIA1. KCNT1 has four cytoplasmic domains, six transmembrane domains, and four extracellular domains. It is widely expressed in the nervous system and its activity contributes to the slow hyperpolarization that follows repetitive firing. KCNT1 is phosphorylated by protein kinase C and phosphorylation of the C-terminal domain is shown to increase its channel activity by 2 to 3-folds. Mutations in Kcnt1 gene have been linked to malignant migrating partial seizures of infancy (MMPSI), a rare epileptic encephalopathy of infancy. Two isoforms of KCNT1 have been described that are produced by alternative splicing. (Ref.: Barcia, G., et al. (2012). Nat. Gen. 44(11); 1255-1259).
Specificity
Clone N3/26 is a mouse monoclonal antibody that detects Potassium channel subfamily member 1 (KCNT1). It targets an epitope within 70 amino acids from the C-terminal region.
Immunogen
Fusion protein corresponding to 70 amino acids from the C-terminal, cytoplasmic domain of rat Potassium channel subfamily member 1 (KCNT1).
Application
Quality Control Testing
Evaluated by Western Blotting in rat brain tissue lysate.
Western Blotting Analysis: A 1:500 dilution of this antibody detected KCNT1 in rat brain tissue lysate.
Tested Applications
Immunohistochemistry (Paraffin) Analysis: A 1:50 dilution from a representative lot detected KCNT1 in rat brain tissue sections.
