Specificity
Clone 1O19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Histone H2A. It targets an epitope within 12 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 12 amino acids from the C-terminal half of yeast Histone H2A.
Application
Quality Control Testing
Evaluated by Western Blotting in Yeast cell acid extract.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Histone H2A in Yeast cell acid extract.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Histone H2A in HepG2 cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound Yeast Histone H2A peptide with a KD of 9.7 x 10-8 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Histone H2A in Human cerebral cortex tissue sections.

Target description
Histone H2A.1 (UniProt: P04911; also known as H2A, H2A.1) is encoded by the HTA1 (also known as H2A1, SPT11, YD9934.10, YDR225W) gene (Gene ID: 851811) in yeast (Saccharomyces cerevisiae). Histones are highly conserved basic NuCLEAR™ proteins that serve as the structural scaffold for the organization of NuCLEAR™ DNA into chromatin. They play a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. These post-translational modifications, in response to various stimuli, induce changes in the structure of the nucleosome and hide or expose DNA sequences. Histone H2A can undergoes acetylation on lysine 5,which promotes unfolding of chromatin and transcription. Although H2A is well conserved, however in contrast to vertebrates and insects, the C-terminus of yeast H2A is not monoubiquitinated. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which DNA is wrapped in repeating units known as nucleosomes, which limits DNA accessibility to the cellular machineries that require DNA as a template. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity™, reproducibility, and stability over conventional monoclonals.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
