Specificity
Clone 1M7 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Histone Macro H2A.1. It targets an epitope within 17 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half in isoform 2 of human Histone Macro H2A.1.
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Histone Macro H2A.1 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Histone Macro H2A.1 in Neuro-2a cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Histone Macro H2A.1 in Human cerebral cortex tissue sections.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Histone Macro H2A.1 in HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound Histone Macro H2A1 peptide with a KD of 1.7 x 10-7 in an affinity binding assay.

Target description
Core histone macro-H2A.1 (UniProt: O75367; also known as Histone macroH2A1, mH2A1, Histone H2A.y, H2A/y, Medulloblastoma antigen MU-MB-50.205) is encoded by the MACROH2A1 (also known as H2AFY) gene (Gene ID: 9555) in human. Histone Macro H2A.1 (mH2A1) is a variant of histone H2A that replaces conventional H2A in a subset of nucleosomes where it represses transcription. Three isoforms of Histone Macro H2A.1 have been described that are produced by alternative splicing. Isoform 1 specifically binds poly-ADP-ribose and O-acetyl-ADP-ribose and plays a KEY® role in NAD+ metabolism. It is able to bind to the ends of poly-ADP-ribose chains created by PARP1 and cap them. This prevents PARP1 from further addition of ADP-ribose and thus limits the consumption of NuCLEAR™ NAD+, allowing the cell to maintain proper NAD+ levels in both the nucleus and the mitochondria to promote proper mitochondrial respiration. Isoform 2 does not bind poly-ADP-ribose. The macro domain of mH2A1 that resides in residues 184-367 is responsible for poly-ADP-ribose binding. mH2A1 is reported to inhibit the binding of transcription factors, including NF-kB, and interfere with the activity of remodeling SWI/SNF complexes. It also inhibits histone acetylation by EP300 and can recruit class I histone deacetylases that induces a hypoacetylated state of chromatin. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
