Specificity

Clone 1E5 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Hepatocyte nuclear factor 3a (HNF-3A/FOXA1). It targets an epitope within 14 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal region of human Hepatocyte nuclear factor 3a (HNF-3A/FOXA1).

Application

Quality Control Testing

Evaluated by Western Blotting in HepG2 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected HNF-3A/FOXA1 in HepG2 cell lysate.

Tested applications

Flow Cytometry Analysis: 0.1 μg from a representative lot detected HNF-3A/FOXA1 in one million HepG2 cells.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected HNF-3A/FOXA1 in HepG2 and MCF-7 cells.

Affinity Binding Assay: A representative lot of this antibody bound HNF-3A/FOXA1 with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HNF-3A/FOXA1 in human prostate tissue sections.

Target description

Hepatocyte nuclear factor 3-alpha (UniProt: P55317; also known as HNF-3-alpha, HNF-3A, Forkhead box protein A1, Transcription factor 3A, TCF-3A) is encoded by the FOXA1 (also known as HNF3A, TCF3A) gene (Gene ID: 3169) in human. HNF-3A is a transcription factor that is involved in embryonic development, establishment of tissue-specific gene expression, and regulation of gene expression in differentiated tissues. It acts as a ′pioneer′ factor opening the compacted chromatin for other proteins through interactions with nucleosomal core histones and thereby replacing linker histones at target enhancer and/or promoter sites. It is also involved in cell cycle regulation by activating expression of p27Kip1 (CDKN1B), alone or in conjunction with BRCA1. In healthy adults, it is expressed in gut endoderm-derived tissues, particularly in the liver. However, high expression of HNF-3A is reported in prostate and estrogen receptor 1-positive breast tumors and in esophageal and lung adenocarcinomas. It binds DNA with the consensus sequence 5′-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3′ and its DNA binding region is localized in amino acids 169-260 (Forkhead). Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
