Specificity

Clone 1B15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects HSC70. It targets an epitope within 20 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal region of human HSC70.

Application

Quality Control Testing

Evaluated by Western Blotting in A431 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HSC70 in A431 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected HSC70 in lysates from L6, HL-60 cells and mouse heart tissue.

Affinity Binding Assay: A representative lot of this antibody bound in HSC70 with a KD of 2.9 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HSC70 in Caco-2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected HSC70 in human testis tissue sections.

Target description

Heat shock cognate 71 kDa protein (UniProt: P11142; also known as EC:3.6.4.10, Heat shock 70 kDa protein 8, Lipopolysaccharide-associated protein 1, LAP-1, LPS-associated protein 1, HSC70) is encoded by the HSPA8 (also known as HSC70, HSP73, HSPA10) gene (Gene ID: 3312) in human. HSC70 is a ubiquitously expressed protein that serves as a molecular chaperone and is implicated in variety of cellular processes, including protection from the proteome stress, folding and transport of newly synthesized polypeptides, activation of proteolysis of misfolded proteins. Hence, it plays a vital role in protein quality control system to ensure proper folding and refolding of misfolded proteins. It also plays a critical role in mitochondrial import and delivers preproteins to the mitochondrial import receptor TOMM70. These functions of HSC70 are achieved through cycles of ATP binding, ATP hydrolysis and ADP release, mediated by co-chaperones. The co-chaperones not only regulate different steps of the ATPase cycle of HSC70, but they also have an individual specificity such that one co-chaperone may promote folding of a substrate while another may promote degradation. Its N-terminal nucleotide binding domain (NBD; aa 2-386) is responsible for binding and hydrolyzing ATP and the C-terminal substrate-binding domain (SBD; aa 394-509) binds to the client/substrate proteins. The two domains are allosterically coupled so that, when ATP is bound to the NBD, the SBD binds relatively weakly to client/substrates. When ADP is bound in the NBD, a conformational change enhances the affinity of the SBD for client/substrate proteins. HSC70 is mainly localized in cytoplasmic mRNP granules containing untranslated mRNAs. However, it translocates rapidly from the cytoplasm to the nuclei upon heat shock. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
