Specificity
Clone 4E12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Heat Shock 70 kDa protein 1A. It targets an epitope within 16 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal region of human Heat Shock 70 kDa protein.
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HSP70 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected HSP70 in MCF-7 and COS-7 cell lysates.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected HSP70 in human bladder tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HSP70 in HeLa, A431, and HepG2 cells.
Affinity Binding Assay: A representative lot of this antibody bound HSP70 peptides with a KD of 3.9 x 10-7 in an affinity binding assay.

Target description
Heat shock 70 kDa protein 1A (UniProt: P0DMV8; also known as Heat shock 70 kDa protein 1, HSP70-1, HSP70.1) is encoded by the HSPA1A (also known as HSP72, HSPA1, HSX70) gene (Gene ID: 3303) in human. The 70-kDa heat shock protein (HSP70) is highly conserved, ubiquitous protein that functions as a molecular chaperone and helps in folding of newly synthesized or denatured proteins and in activation of proteolysis of misfolded proteins and the formation and dissociation of protein complexes. It plays a pivotal role in the protein quality control system, ensuring the correct folding of proteins, the re-folding of misfolded proteins and controlling the targeting of proteins for subsequent degradation. This is achieved through cycles of ATP binding, ATP hydrolysis and ADP release, mediated by co-chaperones. HSP70 is localized in cytoplasmic mRNP granules containing untranslated mRNAs. It is acetylated at Lysine 77 by NA110 in response to cellular stress and then it is gradually deacetylated by HDAC4 at later stages. Its acetylation enhances its chaperone activity and determines whether it will function as a chaperone for protein refolding or degradation by controlling its binding to co-chaperones HOPX and STUB1. The acetylated form and the non-acetylated form are shown to bind to HOPX and STUB1, respectively. HSP70 contains four ATP-binding regions and its N-terminal nucleotide binding domain (NBD; ATPase domain) is responsible for binding and hydrolyzing ATP. Its substrate binding domain (SBD) is localized to the C-terminal region. When ADP is bound in the NBD, a conformational change enhances the affinity of the SBD for client proteins. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
