General description

By incubating any ColorWheel^® antibody with any ColorWheel^® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Specificity

Clone SN6 is a mouse monoclonal antibody that detects Endoglin (CD105). It targets an epitope with in the extracellular domain from the N-terminal half.

Immunogen

Cell membranes of leukemia cells derived from a patient with non-T/non-B-cell acute lymphoblastic leukemia (ALL).

Application

Quality Control Testing:

Evaluated by Flow Cytometry in Human monocytic cell line THP-1.

Flow Cytometry Analysis (FC): Staining of one million Human THP-1 cells was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1060, Anti-Human CD105 (SN6) ColorWheel^® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel^® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Mouse IgG1 isotype control.

Target description

Endoglin (UniProt: P17813; also known as CD105) is encoded by the ENG (also known as END) gene (Gene ID: 2022) in human. CD105 is a disulfide-linked homodimeric, single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 26-586), a transmembrane domain (aa 587-611), and a cytoplasmic domain (aa 612-658). Its expression is observed on endothelial cells and is also detected in placenta. It is also strongly expressed in the angiogenic vasculature of solid tumors. It plays a role in the regulation of angiogenesis and is required for normal structure and integrity of adult vasculature. It regulates the migration of vascular endothelial cells and is shown to be essential for normal extraembryonic angiogenesis and for embryonic heart development. It is reported to play a critical role in the binding of endothelial cells to integrins and/or other RGD receptors. CD105 also acts as TGF-beta coreceptor and participates in the TGF-/BMP signaling cascade that leads to the activation of SMAD transcription factors. Clone SN6 is reported to suppress tumor growth by inhibiting tumor-induced angiogenesis and metastasis. Mutations in ENG gene are reported to cause hereditary hemorrhagic Telangiectasia that is characterized by recurrent epistaxis and gastro-intestinal hemorrhage. (Ref.: She, X., et al. (2004). Int. J. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
