ColorWheelColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheelColorWheel® antibody with any ColorWheelColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheelColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheelColorWheel® technology requires both ColorWheelColorWheel® antibody and ColorWheelColorWheel® dye for Flow Cytometry application.

Specificity
Clone 2E5 is a mouse monoclonal antibody that detects CD253 (TRAIL). It targets an epitope within the extracellular domain.
Immunogen
A recombinant fragment corresponding to 187 amino acids from the extracellular domain of human CD253 (TRAIL).

Target description
Tumor necrosis factor ligand superfamily member 10 (UniProt: P50591; also known as Apo-2 ligand, Apo-2L, TNF-related apoptosis-inducing ligand, Protein TRAIL, CD253) is encoded by the TNFSF10 (also known as APO2L, TRAIL) gene (Gene ID: 8743) in human. TRAIL/CD253 is a single-pass type II membrane protein with a cytoplasmic domain (aa 1-17), a transmembrane domain (aa 18-38), and an extracellular domain (aa 19-281). It is widely expressed and is most predominant in the spleen, lung, and prostate tissue. In addition to the membrane form, a soluble form has also been reported. It exists as a homotrimer where one unit of TRAIL (TNFSF10) interacts with three TRAIL1 (TNFSF10A) or TRAIL2 (TNFSF10B) monomers. TRAIL is a cytokine that binds to TRAIL receptors. Its activity can be modulated by binding to decoy receptors TRAILR3 and TRAILR4. TNFSF10 gene is reported to be a p53-transcriptional gene with two p53 binding sites. Its silencing by siRNA is reported to suppress caspase 3 and caspase 7 activities even after treatment with the DNA damaging agents, such as Adriamycin. TRAIL/CD253 protein expression is shown to be elevated in Adriamycin-treated breast cancer cells. However, it is also shown to engage the apoptosis of tumor cells in a p53 independent manner. Soluble recombinant TRAIL has been shown to induce apoptosis in a wide variety of cancer cells in vitro and to specifically limit tumor growth without damaging normal cells and tissues in vivo. Physical form
Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
Reconstitution
1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
