By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity

Clone UCHT1 is a mouse monoclonal antibody that detects human T-cell surface glycoprotein CD3.

Immunogen

Human infant thymocytes and peripheral blood lymphocytes from a Sézary Syndrome donor.

Flow Cytometry Analysis (FC): Staining of one million Human peripheral blood mononuclear cells (PBMC) was performed using 5 μL of 1:1 mixture of Cat. No. CWA-1019, Anti-Human CD3 (UCHT1) Colorwheel® Dye-Ready mAb and Cat. No. CWD-PE Colorwheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of a PE-conjugated Mouse IgG1 isotype control.

Target description

T-cell surface glycoprotein CD3 epsilon chain (UniProt: P07766; also known as T-cell surface antigen T3/Leu-4 epsilon chain, CD3e) is encoded by the CD3E (also known as T3E) gene (Gene ID: 916) in human. CD3 is a single-pass type I membrane protein that is a part of the TCR-CD3 complex present on T-cells and plays an essential role in adaptive immune response. The TCR CD3 complex is formed through noncovalent association of TCR with a CD3 signalling apparatus consisting of the , , and subunits. It is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 23-126), a transmembrane protein (aa 127-152), and a cytoplasmic domain (aa 153-207). The CD3 complex performs two major functions. Firstly, CD3 chains are required for intracellular signaling where tyrosines in its immunoreceptor tyrosine-based activation motif (ITAM) domain (aa 178-205) are phosphorylated by Lck. Secondly, the CD3 complex is involved in TCR surface expression. In the endoplasmic reticulum, the TCR heterodimer physically associate with the CD3 complex prior to moving to the Golgi for glycosylation and subsequently translocating to the T-cell surface. Mutations in CD3E gene are known to cause immunodeficiency 18 (IMD18) that is characterized by recurrent infections in infancy or early childhood with variable severity. (Ref.: Dong, D., et al. Physical form

Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.

Reconstitution

1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
