General description
ColorWheelColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheelColorWheel® antibody with any ColorWheelColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheelColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheelColorWheel® technology requires both ColorWheelColorWheel® antibody and ColorWheelColorWheel® dye for Flow Cytometry application.

Target description
Integrin alpha-1 (UniProt: P56199; also known as CD49 antigen-like family member A, Laminin and collagen receptor, VLA-1, CD49a) is encoded by the ITGA1 gene (Gene ID: 3672) in human. CD49a/ITGA1 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-28), which is subsequently cleaved off to generate the mature form that contains a large extracellular domain (aa 29-1141), transmembrane domain (aa 1142-1164), and a short cytoplasmic tail (aa 1165-1179). It contains an Integrin I domain (VWFA; aa 161-360) and integrins with I-domain do not undergo proteolytic cleavage. CD49a/ITGA1 forms a heterodimer with integrin ï¢1 that serves as a receptor for laminin and collagen. It recognizes the proline-hydroxylated sequence GFPGER in collagen. CD49a/ITGA1 is shown to be frequently up-regulated in pancreatic ductal adenocarcinoma (PDAC) and associated precursor lesions. It is required for collagen-induced PDAC cell attachment and spreading. Expression of ITGA1-specific collagens within the pancreatic cancer microenvironment significantly correlate with poor patient prognosis. CD49a/ITGA1 levels are also reported to be frequently up-regulated in other cancers, such as melanoma, prostate, bladder, liver. and myeloma. Cd49a/ITGA1 is also required for TGF-β/collagen-induced EMT and metastasis. (Ref.: Gharibi, A., et al. (2017). Sci. Rep. 7; 10060).
Physical form
Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
Reconstitution
1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
