ColorWheelColorWheel® technology is a novel and proprietary method of creating your own antibody and dye combinations for use in flow cytometry. By incubating any ColorWheelColorWheel® antibody with any ColorWheelColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheelColorWheel® antibody and dye is lyophilized for long-term storage, allowing for simplicity and flexibility without compromise. ColorWheelColorWheel® technology requires both ColorWheelColorWheel® antibody and ColorWheelColorWheel® dye for Flow Cytometry application.

Target description
Keratin, type I cytoskeletal 18 (UniProt: P05783; also known as Cell proliferation-inducing gene 46 protein, Cytokeratin-18, CK-18, Keratin-18, K18) is encoded by the KRT18 (also known as CYK18, PIG46) gene (Gene ID: 3875) in human. Cytokeratin-18 (CK-18)is a type I, acidic, intermediate filament protein that is expressed in single-layered epithelial tissues. Its expression is observed in epithelial cells of stomach, duodenum, ileum, colon, placenta, and liver. Higher frequency of CK-18 positivity is observed in adenocarcinomas of the lung, small intestine, prostate, pancreas and the lymph nodes of breast carcinoma. It forms heteropolymers with its co-expressed complementary type II keratin partner CK-8, which assembles into keratin filaments. It is involved in the uptake of thrombin-antithrombin complexes by hepatic cells. Upon phosphorylation, it plays a role in filament reorganization. It is proteolytically cleaved by caspases during epithelial cell apoptosis and necrosis and this cleavage is shown to occur at Asp-238. Phosphorylation of cytokeratin-18 at serine 33 is shown to increase during mitosis in cultured cells and in regenerating liver. Serine 33 phosphorylation is considered to be essential for its association with 14-3-3 proteins and has a role in keratin organization and distribution. Cytokeratin-18 can undergo O-glycosylation, which results in its increased solubility and reduction in its stability. Mutations in KRT18 gene are known to cause liver cirrhosis characterized by severe panlobular liver-cell swelling with Mallory body formation, prominent pericellular fibrosis, and marked deposits of copper. Physical form
Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
Reconstitution
1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL PBS. Protect from light.
