Specificity

Clone 2O9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects IGF-2R/CD222. It targets an epitope within 20 amino acids from the C-terminal half, lumenal domain.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the C-terminal half of human IGF-2R/CD222.

Application

Quality Control Testing

Evaluated by Western Blotting in SK-MEL-28 cell lysates.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IGF-2R/CD222 in SK-MEL-28 cell lysates.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected IGF-2R/CD222 in Jurkat and HepG2 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound IGF-2R/CD222 peptide with a KD of 3.7 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IGF-2R/CD222 in SK-MEL-28 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IGF-2R/CD222 in human placenta tissue sections.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected IGF-2R/CD222 in one million HepG2 cells.

Target description

Cation-independent mannose-6-phosphate receptor (UniProt: P11717; also known as CI Man-6-P receptor, CI-MPR, M6PR, 300 kDa mannose 6-phosphate receptor, MPR 300, Insulin-like growth factor 2 receptor, Insulin-like growth factor II receptor, IGF-II receptor, M6P/IGF2 receptor, M6P/IGF2R, CD222) is encoded by the IGF2R (also known as MPRI) gene (Gene ID: 3482) in human. IGF-2R/CD222 is a single-pass type I membrane protein found in Golgi and endosome. It is synthesized with a signal peptide (aa 1-40), which is subsequently cleaved off to produce the mature protein that contains a long lumenal domain (aa 41-2304), a transmembrane domain (aa 2305-2327), and a cytoplasmic domain (aa 2328-2491). It contains 15 mannose 6-phosphate receptor homology (MRH) domains in its extracellular region. IGF-2R/CD222 mediates the transport of phosphorylated lysosomal enzymes from the Golgi complex and the cell surface to lysosomes. Lysosomal enzymes bearing phosphomannosyl residues bind specifically to mannose-6-phosphate receptors (MPR) in the Golgi apparatus and the resulting receptor-ligand complex is transported to an acidic pre-lysosomal compartment where the low pH mediates the dissociation of the complex. The receptor is then recycled back to the Golgi for another round of trafficking through its binding to the retromer. IGF-2R/CD222 undergoes cysteine-S-palmitoylation that promotes interaction with the retromer cargo-selective complex which mediates its retrograde trafficking to the Golgi apparatus. IGF-2R/CD222 is reported to transport Lck intracellularly and direct its distribution to CD45 at the plasma membrane. The loss of IGF-2R/CD222 results in a decrease in signal transduction and subsequently a blockade of T cell effector functions. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
