Specificity
Clone 1C18 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects JNK1. It targets an epitope within 16 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 16 amino acids from the C-terminal region of human Stress-activated protein kinase JNK1.
Application
Quality Control Testing
Evaluated by Western Blotting in A431 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected JNK1 in A431 cell lysate.
Tested applications
Western Blotting Analysis: A 1:10,000 dilution of a representative lot detected JNK1 in MCF-7 and SH-SY5Y lysates.
Affinity Binding Assay: A representative lot of this antibody bound JNK1 peptide with a KD of 8.5 x 10-6 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected JNK1 in SH-SY5Y cella.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected JNK1 in human thyroid tissue sections.

Target description
Mitogen-activated protein kinase 8 (UniProt: P45983; also known as EC:2.7.11.24, MAP kinase 8, MAPK 8, JNK-46, Stress-activated protein kinase 1c, SAPK1c, Stress-activated protein kinase JNK1, c-Jun N-terminal kinase 1) is encoded by the JNK1 (also known as MAPK8, PRKM8, SAPK1, SAPK1C) gene (Gene ID: 5599) in human. SAPK1/JNK1 is a serine/threonine-protein kinase that is involved in various biological processes, including cell proliferation, differentiation, migration, transformation, and programmed cell death. It is activated in response to extracellular stimuli such as proinflammatory cytokines or physical stress. Both JNK1 and JNK2 contain a TXY motif (aa 183-185) that contains the threonine and tyrosine residues whose phosphorylation activates these kinases. Its protein kinase domain of is localized to amino acids 26-321. Phosphorylation and activation of JNK1 is achieved by two dual specificity kinases, MAP2K4 and MAP2K7. MAP2K4 displays strong preference for Tyr 185 and MAP2K7 phosphorylates Tyr 183. Phosphorylated form of JNK1 is reported to be more concentrated at synapses. Following its activation, JNK1 phosphorylate several transcription factors, primarily components of AP-1 such as JUN, JDP2, and ATF2 and thus regulates AP-1 transcriptional activity. Five isoforms of JNK1 have been described that are produced by alternative splicing. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
