Specificity
Clone 4O11 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects L-Amino acid oxidase. It targets an epitope with 22 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human L-Amino acid oxidase.
Application
Anti-L-amino acid oxidase, clone 4O11 ZooMAb, Cat. No. ZRB1022, is a recombinant Rabbit monoclonal antibody that targets L-Amino acid oxidase and is tested in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry, and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected L-amino acid oxidase in lysates from human colon tissue, THP-1 cells, rat spleen, and mouse spleen tissues.
Affinity Binding Assay: A representative lot of this antibody bound L-Amino Acid Oxidase peptide with a KD of 7.0 x 10-9 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: 1:100 from a representative lot detected L-amino acid oxidase in human tonsil tissue sections.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected L-amino acid oxidase in HeLa, A431, HepG2 and NIH 3T3 cells.

Target description
L-amino-acid oxidase (UniProt: Q96RQ9; also known as EC:1.4.3.2, LAAO, LAO, Interleukin-4-induced protein 1, IL4-induced protein 1, Protein Fig-1, hFIG1) is encode by the IL4I1 (also known as FIG1, UNQ636/PRO1265) gene (Gene ID: 259307) in human. LAAO is a flavoenzyme containing, non-covalently bound flavin adenine dinucleotide enzyme. It is widely distributed and catalyzes the stereospecific oxidative deamination of L-amino acids to produce alpha-keto acids and it also produces ammonia and hydrogen peroxide via an imino acid intermediate. The initial step in its action involves removal of two hydrogen atoms by the flavin coenzyme, with formation of an unstable -amino acid intermediate, which undergoes decomposition by addition of water and forms the ammonium ion and the corresponding -keto acid. LAAO has been shown to selectively bind to cell membranes of specific phospholipid composition and deliver the hydrogen peroxide to localized regions on the cell surface thereby inducing local oxidative stress. Common substrates for LAAO include L-isomers of leucine, isoleucine, nor-leucine, alpha-amino butyric acid, phenylalanine, tyrosine, tryptophan norvaline, methionine, histidine, and citrulline. LAAO is believed to contain three hydrophobic substrate binding sites, designated a, b, and c. Subsite a accommodates one methylene carbon, subsite b two, and subsite c accommodates three carbons. It also contains an amino binding subsite, which is designated as the d site. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
