Specificity
Clone 1D17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects LEAP2. It targets an epitope within 14 amino acids from the internal region.
Immunogen
KLH-conjugated linear peptide corresponding to 14 amino acids from the internal region of human LEAP2.
Application
Quality Control Testing
Evaluated by Western Blotting with recombinant Human LEAP2.
Western Blotting Analysis (WB): 1:1,000 dilution of this antibody detected recombinant Human LEAP2.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LEAP2 in human skeletal muscle.
Affinity Binding Assay: A representative lot of this antibody bound LEAP2 peptide with a KD of 7.0 x 10-6 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected LEAP2 in human skeletal muscle tissue sections.

Target description
Liver-expressed antimicrobial peptide 2 (UniProt: Q969E1; also known as LEAP-2) is encoded by the LEAP2 gene (Gene ID: 116842) in human. Cationic antimicrobial peptides are widely distributed in nature and have a key role in innate immunity and host defense and induce the direct destruction of microorganisms. LEAP2 is a 40-residue cationic antimicrobial peptide that is highly conserved in mammals. LEAP-2 is mainly expressed in the liver but is also produced in smaller quantities in other tissues. It is synthesized with a signal peptide (aa 1-22) and a propeptide (aa 23-37) that are subsequently cleaved off to generate the active mature peptide (aa 38-77). Its primary structure and disulfide bond pattern does not exhibit any similarity to any known peptide. It has a compact central core with disorder at the N- and C-termini. The core comprises a β-hairpin and a 3(10)- helix that are braced by disulfide bonds between Cys17-28 and Cys23-33 (in mature peptide) and is further stabilized by a network of hydrogen bonds. Its expression appears to be well regulated at the transcriptional level. LEAP2 gene consists of three exons and two introns. LEAP2 exhibits dose-dependent antimicrobial activity with an IC50 of about 5 μM against Saccharomyces cerevisiae and Bacillus subtilis. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
