Specificity

Clone 1C17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Leucine-rich repeat serine/threonine-protein kinase 2 (LRRK2). It targets an epitope within 25 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the C-terminal region of human Leucine-rich repeat serine/threonine-protein kinase 2 (LRRK2).

Application

Quality Control Testing

Evaluated by Western Blotting in Human kidney tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected LRRK2/PARK8 in Human kidney tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LRRK2/PARK8 in Mouse brain tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound LRRK2/PARK8 peptide with a KD of 8.1 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LRRK2/PARK8 in A549 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected LRRK2/PARK8 in human kidney and human Parkinson′s diseased brain tissue sections.

Target description

Leucine-rich repeat serine/threonine-protein kinase 2 (UniProt: Q5S007; also known as EC:2.7.11.1, Dardarin, LRRK2) is encoded by the LRRK2 (also known as PARK8) gene (Gene ID: 120892) in human. LRRK2 is a homodimeric serine/threonine protein kinase that phosphorylates a broad range of proteins involved in neuronal plasticity, autophagy, and vesicle trafficking. It is expressed in pyramidal neurons in all cortical laminae of the visual cortex, in neurons of the substantia nigra pars compacta, and caudate putamen. It is expressed throughout the adult brain and its expression is particularly high in dopaminoceptive areas of the brain. LRRK2 acts as a key regulator of Rab GTPases. It regulates the GTP/GDP exchange and interaction partners of RABS through phosphorylation. LRRK2 undergoes autophosphorylation and its phosphorylation on serine 910 and either serine 935 or 1444 facilitates its interaction with tyrosine 3-monooxygenase (YWHAG). LRRK2 is also reported to activate a calcium-dependent protein kinase kinase-β (CaMKK-β)/AMPK pathway, which is followed by a persistent increase in autophagosome formation. Overexpression of LRKR2 increases the levels of the autophagy receptor p62 and reduces the number of acidic lysosomes. Mutations in the LRRK2 gene are known to cause autosomal dominant late-onset Parkinson s disease. Overexpression of LRRK2 is reported to increase phosphorylation of amyloid precursor protein (APP) at a residue within the APP intracellular domain (AICD), which promotes AICD transcriptional activity and neurotoxicity in Parkinson s disease leading to a reduction in dopaminergic neurons. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
