Specificity
Clone 1D11 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Mitogen-activated protein kinase kinase kinase 8 (MAP3K8). It targets an epitope within 17 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal half of human Mitogen-activated protein kinase kinase kinase 8 (MAP3K8).
Application
Quality Control Testing
Evaluated by Western Blotting in L6 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MAP3K8 in L6 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MAP3K8 in lysate from A549 cells and 1:10,000 dilution in lysates from NIH3T3, and HeLa cells.
Affinity Binding Assay: A representative lot of this antibody bound MAP3K8 peptide with a KD of 1.8 x 10-7 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected MAP3K8 in Human brain, Rat brain and Human liver tissue sections.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Target description
Mitogen-activated protein kinase kinase kinase 8 (UniProt: P41279; also known as EC:2.7.11.25, Cancer Osaka thyroid oncogene, Proto-oncogene c-Cot, Serine/threonine-protein kinase cot, Tumor progression locus 2, TPL-2) is encoded by the MAP3K8 (also known as COT, ESTF) gene (Gene ID: 1326) in human. MAP3K8 is involved in the regulation of MAPK cascades and immune responses. Its differential expression and dysregulation are closely correlated with tumorigenesis. It is expressed in several normal tissues and human tumor-derived cell lines. Two isoforms of MAP3K8 have been described that are produced by alternative splicing. It is shown to be involved in LPS-induced, TLR4-mediated activation of the MAPK/ERK pathway in macrophages. It is critical for production of the TNF-α during immune responses. It is also involved in the regulation of T-helper cell differentiation and IFN-γ expression in T-cells. MAP3K8 undergoes autophosphorylation: isoform 1 undergoes phosphorylation mainly on serine residues whereas isoform 2 undergoes phosphorylation on both serine and threonine residues. Its phosphorylation on threonine 290 is necessary but is not sufficient for its full kinase activity. Its phosphorylation on serine 400 by IKK-β is require or LPS-stimulated activation of MAPK pathway in macrophages. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
