General description

MAPKAPK5 (mitogen-activated protein kinase-activated protein kinase 5) is a Ser/Thr kinase, which belongs to a family of MAPK-phosphorylated kinases called MAPKAPK. In humans, eleven such MAPKAPK exist, which are divided into four groups, and MAPKAPK5 belongs to the fourth group, along with MAPKAPK 2 and 3. It was initially recognized as p38 MAKP-activated protein, and is a protein composed of 471 amino acids. It has a ubiquitous expression pattern, and resides primarily in the cytoplasm. This protein shuttles between the cytoplasm and nucleus, and thus, supposedly contains both nuclear localization sequence (NLS) and nuclear export sequence (NES).

Biochem/physiol Actions

MAPKAPK5 (mitogen-activated protein kinase-activated protein kinase 5) is acted upon by ERK (extracellular signal regulated kinase) 3, ERK4 and mitogen-activated protein kinases p38. It acts as a tumor suppressor gene or as an oncogene, and controls cell locomotion, cell cycle control, embryo development and anxiety. It is activated by the presence of microbial pathogen, to elicit an immune response. It is an interacting partner of DJ-1, which is a mitogen-dependent oncogene product. Induction of DJ-1 leads to the accumulation of Daxx in the nucleus, thereby preventing cell apoptosis under oxidative stress. It is involved in doxorubicin-resistance in hepatocellular carcinoma cells, where it is involved in suppressing doxorubicin-induced apoptosis. Doxorubicin also induces the degradation of this protein. This protein interacts with and phosphorylates heat shock protein 27 (Hsp27), which in turn mediates PKA (protein kinase A)-induced rearrangement of F-actin.
