Specificity

Clone 1G13 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects MARC1. It targets an epitope within 16 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 16 amino acids from the internal region, cytoplasmic domain of human Mitochondrial amidoxime-reducing component 1 (MARC1).

Application

Quality Control Testing

Evaluated by Western Blotting in Human liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MARC1 in Human liver tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected MARC1 in Mouse liver and Rat liver tissue lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MARC1 in HepG2 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MARC1 in human liver tissue sections.

Target description

Mitochondrial amidoxime-reducing component 1 (UniProt: Q5VT66; also known as mARC1, Molybdenum cofactor sulfurase C-terminal domain-containing protein 1, MOSC domain-containing protein 1, Moco sulfurase C-terminal domain-containing protein 1) is encoded by the MTARC1 (also known as MARC1, MOSC1) gene (Gene ID: 64757) in human. MARC1 is a single-pass type II outer mitochondrial membrane protein that consists of a mitochondrial matrix domain (aa 2-20), a transmembrane domain (aa 21-40), and a cytoplasmic domain (aa 41-337). It catalyzes the reduction of N-oxygenated molecules, acting as a counterpart of cytochrome P450 and flavin-containing monooxygenases in metabolic cycles. It is known to reduce a multitude of N-hydroxylated prodrugs, particularly amidoximes, leading to increased drug bioavailability. It is also reported to be involved in mitochondrial N(omega)-hydroxy-L-arginine (NOHA) reduction, thereby regulating endogenous nitric oxide levels and biosynthesis. It has two structural domains, the molybdenum cofactor(Moco) sulfurase C-terminal (MOSC) domain (aa 187-335) and the MOSC N-terminal region (aa 93-183), forming a cleft that accommodates Moco. The MOSC domain that contains a large seven-stranded mostly antiparallel beta-barrel, engages multiple interactions with Moco pterin ring and phosphate group, which allows for a tight coordination of Moco within the core of the enzyme. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
