Specificity
Clone 1L20 is a Rabbit recombinant monoclonal antibody that specifically detects Methyl-CpG-binding protein 2 (MeCP2). It targets an epitope within 14 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal region of human Methyl-CpG-binding protein 2 (MeCP2).
Application
Quality Control Testing
Evaluated by Western Blotting in Cos-7 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MeCP2 in Cos-7 cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MeCP2 in MCF-7 and HeLa cell lysates.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MeCP2 in A431, HeLa, HepG2, and NIH 3T3 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected MeCP2 in human liver tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound MeCP2 with a KD of 9.6 x 10-9 in an affinity binding assay.

Target description
Methyl-CpG-binding protein 2 (UniProt: P51608; also known as MeCp-2 protein, MeCP2) is encoded by the MECP2 gene (Gene ID: 4204) in human. MeCP2 is a chromosomal protein that binds to methylated DNA and is present in high abundance and frequency at its target sites. It can bind specifically to a single methyl-CpG pair and is considered as a reader of the DNA methylome. It is shown to binds both 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC)-containing DNA, with a preference for 5-methylcytosine (5mC). It contains a methyl-CpG binding domain (MBD; aa 90-162) that is responsible for its binding to DNA. MeCP2 mediates transcriptional repression through interaction with histone deacetylase and the corepressor SIN3A. It is expressed in all tissues, but higher levels are present in neurons. Its levels increase during embryonic and postnatal development and then plateau off around the age of 10 years in humans. It can undergo multiple post-translational modifications at multiple sites by phosphorylation, ubiquitination, SUMOylation, acetylation, methylation, GlcNAcylation and poly(ADP-ribosyl)lations. These modifications affect its stability and functions. It is reported to be phosphorylated on serine 423 in brain upon synaptic activity that attenuates its repressor activity and regulates dendritic growth and spine maturation. Mutations leading to loss-of-function are known to cause Rett syndrome that leads to mental retardation. Most of these mutations have been found in the MBD region. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
