Specificity
Clone 1F18 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects MEK1. It targets an epitope within 17 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal region of Human MEK1.
Application
Quality Control Testing
Evaluated by Western Blotting in COS-7 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MEK1 in COS-7 cell lysate.
Tested applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MEK1 in HEK293 and U87MG cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound MEK1 peptide with a KD of 3.3 x 10-7 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MEK1 in HEK293 cells.

Target description
Dual specificity mitogen-activated protein kinase kinase 1 (UniProt: Q02750; also known as EC:2.7.12.2, MAP kinase kinase 1, MAPKK 1, MKK1, ERK activator kinase 1, MAPK/ERK kinase 1, MEK 1) is encoded by the MEK1 (also known as MAP2K1, PRKMK1) gene (Gene ID: 5604) in human. MEK1 is a dual specificity protein kinase that acts as an essential component of the MAP kinase signal transduction pathway. It is widely expressed, but very low levels are detected in the brain tissue. Activation of MEK1 involves binding of extracellular ligands such as growth factors and cytokines to their membrane receptors that activates RAS and then RAF. Once activated, GTP-bound RAS can drive the formation of high-activity homodimers or heterodimers of the RAF protein kinases (ARAF, BRAF or CRAF) that can directly phosphorylate and activate MEK1 and MEK2. Their activation occurs through phosphorylation of Ser 218 and Ser 222 residues. MEK1/2 in turn can activate ERK1 and ERK2 by phosphorylating them at conserved threonine and tyrosine residues in the Thr-Glu-Tyr motif in their activation loop. MEK1 is reported to be the target of negative feed-back regulation by its substrate kinases ERK2, which can phosphorylate MEK1 on Thr 292 and facilitate dephosphorylation of the activating residues Ser 218 and Ser 222. Although MEK1/2 mutations are relatively rare in cancer, expression of constitutively active forms of their mutants are reported to be sufficient to induce oncogenic transformation of normal cells. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
