Target description
Tumor necrosis factor ligand superfamily member 13B (UniProt: Q9WU72; also known as B-cell-activating factor, BAFF, CD257) is encoded by the Tnfsf13b (also known as Baff) gene (Gene ID: 24099) in murine species. BAFF is a single-pass type II, homotrimeric, membrane glycoprotein with a cytoplasmic domain (aa 1-47), a transmembrane domain (aa 48-68), and an extracellular domain (aa 69-309). A soluble form of BAFF has also been described that is produced by the cleavage of membrane form between amino acids 126-127. It is expressed by cells of myeloid origin and by stromal cells. Two isoforms of BAFF have been described that are produced by alternative splicing. Isoform 2 is reported to inhibit the secretion and bioactivity of isoform 1. BAFF is reported to bind to TNFRSF13B/TACI and TNFRSF17/BCMA. It serves as a potent B cell survival factor and is a critical immune checkpoint during peripheral B cell maturation. Its levels are often elevated in autoimmune diseases, such as rheumatoid arthritis and systemic lupus erythematosus (SLE). Elevated levels of BAFF are also observed in subjects with non-Hodgkin s lymphoma (NHL) and it is reported to promote survival of both NHL-derived B lymphoma cells and B cell chronic lymphocytic leukemia (B-CLL) cells. Storage and Stability
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
