Specificity
Clone 1O8 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human Methyltransferase-like protein 7A (METTL7A). It targets an epitope within 21 amino acids from the C-terminal region and detects both isoforms.

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human Methyltransferase-like protein 7A (METTL7A).
Application
Quality Control Testing
Evaluated by Western Blotting in HT-1376 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Methyltransferase-like protein 7A (METTL7A) in HT-1376 cell lysate.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound METTL7A peptide with a KD of 1.1 x 10-7 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected METTL7A in human testis tissue sections.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected METTL7A in HepG2 cells.

Target description
Methyltransferase-like protein 7A (UniProt: Q9H8H3 and computationally mapped F8VQX6, Q9H8H3, also known as AAM-B, METTL7A) is encoded by the METTL7A gene (Gene ID: 25840) in human. METTL7A is a ubiquitously expressed lipid droplets-associated protein that displays high expression in liver and thyroid tissues. It is synthesized with a signal peptide (aa 1-29) and is an integral membrane protein anchored into the endoplasmic reticulum (ER) membrane. It contains N and C-terminal domains, juxtamembrane region, and a putative central catalytic domain for methyltransferase activity. The N-terminal regions contains a hydrophobic sequence of 28 amino acids that is shown to be essential for its insertion into ER. It also helps recruit other cellular protein to lipid droplets for the formation of functional organelles. METTL7A is reported to be essential for the assembly and release stages of hepatitis C (HCV) life cycle but is not essential for the replication step of virus. Knockdown of METTL7A is shown to impair HCV propagation. METTL7A gene is reported to be downregulated in thyroid cancer. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
