Specificity
Clone 3L17 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Myeloid/lymphoid or mixed-lineage leukemia protein 2 (MLL2). It targets an epitope within 22 amino acids from the C-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal region of human Myeloid/lymphoid or mixed-lineage leukemia protein 2 (MLL2).
Application
Quality Control Testing
Evaluated by Immunohistochemistry (Paraffin) in human placenta tissue sections.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution of this antibody detected MLL2 in human placenta tissue sections.
Tested Applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MLL2 in U2OS cells.
Affinity Binding Assay: A representative lot of this antibody bound MLL2 C-terminal peptide with a KD of 3.8 x 10-8 in an affinity binding assay.

Target description
Histone-lysine N-methyltransferase 2D (UniProt O14686; also known as EC 2.1.1.43, ALL1-related protein, Lysine N-methyltransferase 2D, Myeloid/lymphoid or mixed-lineage leukemia protein 2, MLL2) is encoded by the KMT2D (also known as ALR, KABUK1, MLL2, MLL4) gene (Gene ID: 8085) in human. MLL2 is a histone methyltransferase that catalyzes methyl group transfer from S-adenosyl-L-methionine to the -amino group of lysine 4 of histone H3 (H3K4) that represents a specific tag for epigenetic transcriptional activation. Its expression is observed in most adult tissues, including a variety of hematopoietic cells. However, it has not been detected in the liver. It predominantly forms H3K4me1 methylation marks at active chromatin sites where transcription and DNA repair take place. It forms a core complex with the evolutionary conserved subcomplex WRAD that is composed of WDR5, RBBP5, ASH2L/ASH2 and DPY30 subunits. Mutations in KMT2D gene have been linked to Kabuki syndrome 1, a congenital mental disorder with multiple abnormalities, including dwarfism, scoliosis, and radiographic abnormalities of the vertebrae, hands, and hip joints. Inactivating mutations in MLL2 gene have been linked to medulloblastoma. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
