By incubating any ColorWheel® antibody with any ColorWheel® dye, researchers can quickly and simply produce primary conjugated antibodies for use in single color or multicolor flow cytometry analysis. Each ColorWheel® antibody and dye is lyophilized for long long-term storage, allowing for simplicity and flexibility without compromise. ColorWheel® technology requires both ColorWheel® antibody and ColorWheel® dye for Flow Cytometry application.

Specificity
Clone 90 is a rat monoclonal antibody that detects mouse ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1 (CD28). It targets an epitope within the C-terminal region.

Mouse bone marrow pre-B cells.
Application
Quality Control Testing:
Evaluated by Flow Cytometry in mouse splenocytes.
Flow Cytometry Analysis: Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1038, Anti-Mouse CD38 (90) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® activated Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2a isotype control.

Target description
ADP-ribosyl cyclase/cyclic ADP-ribose hydrolase 1 (UniProt: P56528; also known as EC:3.2.2.6, 2′-phospho-ADP-ribosyl cyclase, 2′-phospho-ADP-ribosyl cyclase/2′-phospho-cyclic-ADP-ribose transferase, 2′-phospho-cyclic-ADP-ribose transferase, ADP-ribosyl cyclase 1, ADPRC 1, Cyclic ADP-ribose hydrolase 1, cADPr hydrolase 1, I-19, NIM-R5 antigen, CD38) is encoded by the Cd38 gene (Gene ID: 12494) in murine species. CD38, a member of the ADP-ribosyl cyclase family, is a single-pass type II membrane glycoprotein that contains a short cytoplasmic domain (aa 1-21), a transmembrane domain (aa 22-44), and a long extracellular domain (aa 45-304). It is expressed in many immune cell types, including bone marrow progenitors, natural killer cells, monocytes, and activated T- and B- lymphocytes. It serves as a lymphocyte receptor and is a considered to be a prognostic marker for survival with B-cell chronic lymphoblastic leukemia. The catalytic activity of CD38 resides in the extracellular domain that can both generate and degrade cyclic-ADP ribose and can catalyze the base-exchange reaction between NADP+ and nicotinic acid to produce NAADP+. Conformational change in CD38 can influence its catalytic activities. Clone 90 is reported to induce conformational change in CD38 and serves as agonistic antibody. It stimulates NAD+ glycohydrolase activity of FLAG-CD38 but has no effect on its cyclase activity. However, it inhibits the cyclase activity of cell surface CD38. (Ref.: Hartman, WR., et al. (2010). Physical form
Lyophilized from PBS containing D-Mannitol and Sucrose, normal appearance is a dried pellet. Reconstituted antibody solution is stable and functional as assessed by functional testing. Contains no biocide or preservatives, such as azide.
Reconstitution
1.0 mg/mL after reconstitution at 25 μL in PBS. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
