General description
Metallophosphoesterase 1 (MPPE1) belongs to the dimetal-containing phosphoesterase family. It is located at the endoplasmic reticulum-exit site, endoplasmic reticulum-Golgi intermediate compartment and the cis-Golgi. The gene encoding this protein is present on chromosome 18p11.2. It contains a signal sequence of 45 residues at the amino-terminus, a metallo-phosphoesterase domain and a transmembrane domain at the carboxyl-terminus.

Biochem/physiol Actions
Metallophosphoesterase 1 (MPPE1) transports glycophosphatidyloinositol (GPI)-anchored proteins from the endoplasmic reticulum (ER) to the Golgi. This is done by recruiting and sorting them together with the p24 (a transmembrane protein) heteromeric complexes to the ER-exit site. MPPE1 also plays an important role in the lipid remodeling steps of GPI-anchor maturation. It removes the side-chain ethanolamine-phosphate (EtNP) from the second Mannose residue (Man2) of the GPI intermediate. This is one of the crucial steps for the transport of GPI-anchor proteins.
